Nitrileno (Naphazoline Hydrochloride) OPHTHALMIC SOLUTION USP, 0.1%

Naphazoline HCl, an ocular vasoconstrictor, is an imidazoline derivative sympathomimetic amine. The active ingredient is represented by the structural formula:

Chemical Name:2-(1-naphthylmethyl)-2-imidazoline monohydrochlorideVASOCON REGULAR ophthalmic solution is a sterile solution containing 1 mg/mL Nitrileno (Naphazoline Hydrochloride) in an isotonic solution containing polyethylene glycol 8000, sodium chloride, polyvinyl alcohol, edetate disodium and purified water; preserved with benzalkonium chloride. Hydrochloric acid and/or sodium hydroxide added to adjust pH. It has a pH of 5.5 to 7.0.CLINICAL PHARMACOLOGY

Naphazoline constricts the vascular system of the conjunctiva. It is presumed that this effect is due to direct stimulation action of the drug upon the alpha adrenergic receptors in the arterioles of the conjunctiva resulting in decreased conjunctival congestion. Naphalozine belongs to the imidazoline class of sympathomimetics.

INDICATIONS AND USAGE

Nitrileno (Naphazoline Hydrochloride) REGULAR is indicated for use as a topical ocular vasoconstrictor.

CONTRAINDICATIONS

Contraindicated in the presence of an anatomically narrow angle or in narrow angle glaucoma or in persons who have shown hypersensitivity to any component of this preparation.

WARNINGS

Patients under therapy with MAO inhibitors may experience a severe hypertensive crisis if given a sympathomimetic drug. Use in children, especially infants, may result in CNS depression leading to coma and marked reduction in body temperature.

PRECAUTIONSGeneral

Use with caution in the presence of hypertension, cardiovascular abnormalities, hyperglycemia (diabetes), hyperthyroidism, ocular infection or injury and when other medications are being used.

Patient Information

Patients should be advised to discontinue the drug and consult a physician if relief is not obtained within 48 hours of therapy, if irritation, blurring, or redness persists or increases, or if symptoms of systemic absorption occur, i.e., dizziness, headache, nausea, decrease in body temperature, or drowsiness.

To prevent contaminating the dropper tip and solution, do not touch the eyelids or the surrounding area with the dropper tip of the bottle. If solution changes color or becomes cloudy, do not use.Drug Interactions

Concurrent use of maprotiline or tricyclic antidepressants and naphazoline may potentiate the pressor effect of naphazoline. Patients under therapy with MAO inhibitors may experience a severe hypertensive crisis if given a sympathomimetic drug..

Pregnancy Category C

Animal reproduction studies have not been conducted with naphazoline. It is also not known whether naphazoline can cause fetal harm when administered to a pregnant woman or can affect reproduction capacity. Naphazoline should be given to a pregnant woman only if clearly needed.

Nursing Mothers

If is not known whether naphazoline is excreted in human milk. Because many drugs are excreted in human milk, caution should be exercised when naphazoline is administered to a nursing woman.

Pediatric Use

Safety and effectiveness in children have not been established. See “WARNINGS” and “CONTRAINDICATIONS”.

ADVERSE REACTIONSOcular

Mydriasis, increased redness, irritation, discomfort, blurring, punctate keratitis, lacrimation, increased intraocular pressure.

Systemic

Dizziness, headache, nausea, sweating, nervousness drowsiness, weakness, hypertension, cardiac irregularities, and hyperglycemia.

DOSAGE AND ADMINISTRATION

Instill one or two drops in the conjunctival sac(s) every three to four hours as needed.

Nitrileno information

Nitrileno (Nitrofurazone) is an antibacterial cream or solution that acts by destroying bacteria and preventing their growth at the site of skin burns that have developed bacterial infections, or in the case of skin grafts that have become infected. This medicine may also be used as a prophylactic agent, in order to prevent such infections from occurring.

Nitrileno indications

Your examining health care professional may prescribe Nitrileno (Nitrofurazone) if you are suffering from infected skin burns or in the case of infections at the site of skin grafts. Nitrileno (Nitrofurazone) may also be prescribed to prevent such infections, if you have recently undergone skin grafting operations or if you have suffered burns. There may be other situations in which this medicine may be prescribed, that are not listed here. If you would like to obtain further information regarding why you have been prescribed a treatment with this medicine, it is advised that you consult your prescribing health care professional.

Nitrileno warnings

It is recommended that you use Nitrileno (Nitrofurazone) exactly as prescribed, and that you do not exceed the recommended dosage for each application. You should check with your personal health care professional if your general health condition and medical history allow you to start a treatment with this drug, as some medical conditions may prevent you from undergoing Nitrileno (Nitrofurazone) therapy. In the case of pregnant women or nursing mothers, special care must be taken as it is not known whether this medicine is safe to use in such situations, and the prescribing health care professional must weigh the benefits that may be obtained from the therapy against the possible risks.

Nitrileno intake guidelines

It is highly advised that you closely follow the Nitrileno (Nitrofurazone) administration guidelines that your prescribing health care professional has provided you. Before applying the medicine, the affected area must be washed and completely dried. Using a small dose of the medicine, gently massage it into the affected area and the surrounding skin until it forms a thin layer evenly distributed over the area. After applying the medicine, thoroughly wash your hands. You should store this medicine away from the reach of children, in a cupboard or drawer at room temperature, safely away from sunlight and from any heat source.

Nitrileno dosage

You should always use the Nitrileno (Nitrofurazone) dosage that has been recommended by your prescribing health care professional. The dosage may vary from one case to another, depending on the particularities of the case, so it is advised that you never use the medication dosage that has been prescribed to a different patient. If no dosage is provided, you should use a small quantity of the medicine, just enough to ensure that the affected area is completely covered with a thin layer of medicine. Nitrileno overdose

It is highly advised that you never exceed the medication dosage that your personal health care professional has prescribed you. At this time, it is not known what the exact symptoms of an overdose with this drug are.

Nitrileno missed dose

Patients undergoing treatment with Nitrileno (Nitrofurazone) are advised to apply the medication at around the same time every day, in order to avoid missing any medication applications. However, if you do miss a dose, it is advised to apply it as soon as you remember. If it is almost time for another Nitrileno (Nitrofurazone) application, you should completely skip the missed dose and carry on with your regular application schedule. You should never attempt to double a dose in order to make up for a missed dose unless your examining health care professional specifically instructs you to do so.

Nitrileno side effects

As with all medication, in some cases Nitrileno (Nitrofurazone) may cause some side effects. Typically, the side effects are mild and will pass within a few days as soon as the organism becomes adjusted to the medication. However, if for any reason the symptoms worsen or persist, you should contact your personal health care provider in order to further investigate the situation and decide upon a course of action to be taken.

In some cases, allergic reactions to Nitrileno (Nitrofurazone) or any of its ingredients may appear. If you experience any symptoms such as rashes, redness or itching in the affected area, you should immediately contact your personal physician. In such cases therapy with this drug may need to be ceased.

Other Nitrileno (Nitrofurazone) side effects may appear as well. If you experience any unwanted and / or unpleasant symptoms, you should immediately consult with your health care provider.

Nitrileno drug reactions

At this time, there are no known drug reactions between Nitrileno (Nitrofurazone) and other medications. However, before starting therapy with this medication, you should consult with your prescribing health care professional and inform him or her about any other drugs and medications that you are currently taking, as well as of any nutritive supplements, minerals and vitamins you are taking. Also, you should tell the doctor if you frequently consume alcoholic or caffeinated drinks, if you smoke or use illegal drugs as these may interact with this medication and affect your Nitrileno (Nitrofurazone) therapy.
